Localization of Nicotinic Acetylcholine Receptor ¥á7 and ¥â4 Subunits on Direction-Selective Ganglion Cells in Rabbit Retina

Abstract

|
Purpose: We investigated the existence and distribution of the nicotinic acetylcholine receptor (nAChR) ¥á7 and ¥â4 subunits on the dendritic arbors of direction-selective retinal ganglion cells (DSRGCs) in the adult rabbit retina.
Methods: DSRGCs were injected with 5% Lucifer yellow to identify their dendritic morphology. The nAChR ¥á7 and ¥â4 subunits on the dendritic arbors of the DSRGCs in the adult rabbit retina were identified using immunocytochemistry. The double-labeled images of dendrites and nAChR ¥á7 and ¥â4 subunits were visualized for reconstruction using high-resolution confocal microscopy.
Results: The distribution of nAChR ¥á7 and ¥â4 subunits on dendritic arbors in both the ON and OFF layers of the DSRGCs did not reveal an asymmetrical pattern in adult animals.
Conclusions: Although the present study suggests that the distributional pattern of the nAChR ¥á7 and ¥â4 subunits on the dendritic arbors of the DSRGCs was not asymmetric in the adult rabbit retina, the findings provide anatomical evidence of nAChR subunits for direction selectivity of cholinergic input.
